Listeria monocytogenes is a facultative intracellular human and animal pathogen that is commonly isolated from several sources along the human food chain and causes a potentially fatal disease known as listeriosis. Studies were conducted to (i) gain a better understanding of the ecology and transmission of L. monocytogenes in pre-harvest systems, (ii) probe the molecular phylogeny and evolution of L. monocytogenes, and (iii) define genetic mechanisms responsible for differences in the abilities of L. monocytogenes strains to cause disease. A case-control study of listeriosis in ruminants indicated that the epidemiology of listeriosis differs between cattle and small ruminants (goats and sheep). Cattle farms maintain a high L. monocytogenes prevalence and cattle to disperse the organism into the farm environment via fecal shedding thus indicating the cattle farms represent a significant L. monocytogenes reservoir. Molecular epidemiology analyses showed that some L. monocytogenes subtypes may be adapted to cause disease in animals, while others may be adapted to colonize the farm environment. The prevalence of L. monocytogenes on ruminant farms, most notably in fecal samples from healthy animals, was shown to be seasonal and peak during the winter and spring. Risk analysis identified access to pasture, providing quality feedstuffs, and proper feedstuff storage as important farm-management practices to reduce the incidence of ruminant listeriosis and L. monocytogenes fecal shedding. A study on the molecular phylogeny and evolution of L. monocytogenes isolates from humans, animals, and foods showed that L. monocytogenes contains two major deeply separated lineages (I and II) that may represent subspecies or possibly different species. Lineage I appeared to be clonal and possibly host adapted, while lineage II was genetically diverse and may represent a "bacterial generalist" lineage. Three novel non-sense mutations were observed in inlA, the virulence gene encoding internalin A (In1A), a protein that mediates L. monocytogenes invasion of host cells. These inlA mutations resulted in a truncated and secreted InlA and were common among food isolates and rarely associated with human disease. L. monocytogenes isolates with a truncated InlA showed significantly reduced invasion in human intestinal epithelial cells as compared to full-length InlA isolates.
